Phenolic compounds found in Cannabis sativa L., notably the intriguing cannflavins, have long been recognized for their diverse range of biological properties. However, one aspect that has remained relatively unexplored is their potential antiproliferative activity—a facet of immense interest from the standpoint of medicinal chemistry. In this groundbreaking study, researchers from University of Modena and Reggio Emilia embarked on a journey to harness the healing potential of polyphenols extracted from a non-psychoactive C. sativa variety, commonly referred to as hemp. The goal was to obtain an enriched fraction of polyphenols, known as PEF, from hemp inflorescences and rigorously assess their ability to combat cancer cells. To achieve this, the researchers developed an innovative, selective extraction method for hemp polyphenols, followed by preparative flash column chromatography. As a significant advancement in polyphenol purification, this method was instrumental in characterizing these compounds and uncovering their full potential.

The central focus of the study was the assessment of PEF's antiproliferative potential, along with the individual polyphenolic constituents. For the very first time, they conducted in vitro tests on Caco-2 and SW480 human colon adenocarcinoma cell lines. The results were incredibly promising, as PEF and the isolated compounds displayed compelling IC50 values, often outperforming a reference drug commonly used in the treatment of colorectal cancer.

Comprehensive Characterization Using UHPLC-HRMS:

The approach to fully understanding the polyphenolic extract involved a novel method based on ultra-high-performance liquid chromatography coupled with high-resolution mass spectrometry (UHPLC-HRMS). This groundbreaking technique allowed us to gain an in-depth understanding of the PEF, unveiling a plethora of polyphenolic compounds that were previously hidden from view.

Quantification of Key Phenolics with HPLC-UV

Simultaneously, the researchers quantified the primary phenolic compounds within the PEF using high-performance liquid chromatography with ultraviolet detection (HPLC-UV). This quantification not only provided insights into the concentration of essential polyphenols but also offered a foundation for understanding their potential medicinal benefits.
